Package #3: Custom Monospecific Antibody Production

Overview:

Custom Monospecific Antibody Production is our most popular package. It is designed for generating affinity-purified antibodies against a synthesized peptide.

Antigen Design:

Pacific Immunology provides antigen design assistance at no additional charge. Please send your sequence(s) to our Sales team with any details on regions to target/avoid, and we will be happy to send our recommendations. Once the final sequence(s) has been chosen, we can then put together a formal quotation for the project.

Conjugation of Peptide to Carrier Protein:

Because peptides are too small to generate an immune response by themselves, conjugation to a carrier protein will be required for immunizations. Pacific Immunology recommends coupling peptides to KLH (Keyhole Limpet Hemocyanin).

Terminal Cysteine:

Pacific Immunology recommends coupling the peptide to the carrier protein via a cysteine that has been added to the N or C-terminal of the peptide. For this reason, we recommend avoiding sequences that contain internal cysteines whenever possible.

Quantity of Peptide:

Pacific Immunology typically synthesizes 40-50 mg of immunograde peptide, with approximately 10 mg being required for conjugation to the carrier protein, ELISA and affinity purification. All unused peptide will be delivered to the customer.

Affinity Column:

The affinity column is included so that the client can run additional purifications as needed. Most columns can be reused at least 10 times within a 6-12 month period.

Titer Guarantees:

Pacific Immunology offers one of the highest titer guarantees in the industry, promising at least a 1:20,000 antibody titer against the peptide sequence.

Purified Antibody Yields:

On average, 2-3 mg of peptide specific antibody will be isolated from affinity purification of 25 ml of antiserum.

Affinity Purification Strategy:

Unless one animal has particularly high antibody titers against the peptide, we recommend affinity-purifying a pool of 25 ml serum taken in a 1:1 ratio from both animals to ensure maximum epitope variability. Comprehensive ELISA testing confirms the success of the purification.

Comparison to Monoclonal Antibodies:

Monospecific antibodies offer significant price and timeline advantages in comparison to monoclonal antibodies, while offering similar specificity. With large-scale purifications, very large yields of purified antibody can be obtained.
